RESUMO

Anilino naphthalenesulfonate fluorescence dynamics in the rat liver mitochondria during the first six minutes after corticosteroid (corticosterone, cortisone, hydrocortisone, dexamethasone) and cholesterol treatment in vitro was studied. The biphasic character of the process was observed depending on the hydrophobicity and concentration of the steroids.

RESUMO

Redox-reactions of flavine (FN) and pyridine (PN) nucleotides of the cell to guthimine were studied by the method of fluorescent microscopy without using any stains. The comparison was made of the kinetics of fluorescence intensity of FN and PN of the Retzius intact neuron under the action of guthimine, oxaloacetate, nembutal and succinate. It is suggested that guthimine may activate the switching of oxidative metabolism of the cell over to a more energitically profitable way.

RESUMO

The influence of corticosteroid hormones and cholesterol on the anilinonaphthalenesulfonate (ANS-) fluorescence in the rat liver mitochondria at different temperatures has been studied. The fluorescence intensity has been found to decrease by warming and increase by cooling the mitochondrial suspension. The steroids studied in proportion to their hydrophobic nature decreased the ANS- fluorescence at 25 degrees C and increased it at 5 degrees C, i.e. at higher and lower temperatures of the phase transition of the membrane lipids. It has been concluded that steroid hormones influence the fluidity of the mitochondrial membranes.

RESUMO

It has been shown on Retzius neuron of the leech that after preintroduction of sodium succinate the reaction to synaptic stimulation expressed in increased frequency of the impulse activity proceeds more intensively than in the norm. In connection with the fact that against the background of succinate effect reaction to acetylcholine increases it is suggested that this very mediator is responsible for slower decrease of the impulse activity frequency at synaptic activation.

RESUMO

It has been shown on leech Retzius neuron that the effect of succinic acid on impulse activity, respiration and redox state of flavin nucleotides depends on initial values of these parameters with the exception of membrane potential. In this case independent of its initial value the membrane potential becomes equal to 35 +/- 6 mV. The analysis of the evidence obtained has shown that the higher is the activation of cell respiration under the effect of succinic acid, the less are changes in redox-reaction of flavin nucleotides. At stronger activation of respiration the frequency of impulse activity is increased and the value of membrane potential decreased. At smaller changes of respiration the impulse activity frequency is not changed while the membrane potential increases as a rule.

RESUMO

On different Retzius cell preparations (cell in the whole leech, in part of the animal and in isolated ganglionic chain) it has been demonstrated that the process of preparation itself affects redox levels of flavoproteins and pyridine nucleotides in the intact cell, changes their redox response to oxygen and alters the direction of redox reaction of pyridine nucleotide to 5 mM oxaloacetate. Redox levels of flavin and pyridine nucleotides in the cell were evaluated by the intensity of its fluorescense in violet and green parts of the spectrum.
